μCaler RIA Solution is built on EZ RNA & DNA Library Co-Preparation kit, combined with the μCaler Hybrid System, μCaler RIA Panel v1.0, and the XCapViz Bioinformatics Analysis Visual System. Together, they deliver an integrated workﬂow spanning sample processing, library preparation, hybrid capture, bioinformatics analysis, and report interpretation, enabling a stream-lined “sample-in, result-out” process. In addition, the ﬂexible NadAuto-series Fully Automated NGS Workstations can further reduce hands-on labor and increase throughput and eﬃciency for clinical respiratory pathogen testing.

μCaler RIA Panel v1.0 is designed to target speciﬁc genomic sequences from hundreds of respiratory pathogens, as well as ~200 AMR markers associated with commonly used antimicrobials. With coverage of > 99% of common clinically relevant respiratory pathogens, it enables precise pathogen identiﬁcation and comprehensive resistance proﬁling, providing strong technical support for rapid diﬀerentiation of complex infections, accurate diagnosis, individualized therapy decisions, and ongo-ing surveillance and management.

Application

Upper respiratory tract infections (URTIs): common cold, influenza, rhinitis, pharyngitis, tonsillitis, laryngitis, etc.

Performance

Stable Detection of Multiple Pathogens in Reference Standard

A total of 0.4 mL of reference standard at diﬀerent concentration gradients was used for extraction of mixed RNA & DNA. Pre-libraries were prepared using the NadPrep EZ RNA & DNA Library Co-Preparation Module coupled with the NadPrep Universal Stubby Adapter (UDI) Module. Hybrid capture was performed using the μCaler HyperMix Hybrid Capture Kit (for Illumina®) and the μCaler RIA Panel v1.0. Sequencing was performed on MiSeq with PE150, with raw data of 3 M read pairs per sample.

Note: RPM stands for reads per million mapped reads.

Possible Reason	Solution
Input amount of nucleic acid is too low	If the sample is a co-extracted total nucleic acid, quantiﬁcation of RNA and DNA should be performed separately using Qubit, and library ampliﬁcation should be carried out according to the recommended cycles in the manual.
	During the experiment, RNase-free consumables must be used, and the operating environment should be free of RNase contamination, as this can aﬀect the eﬃciency of RNA reverse transcription.
	If the nucleic acid sample contains excessive amounts of chelating agents, guanidine salts, phenol, proteins, ethanol, or other impurities, it can impact the activity of RNA reverse transcriptase and the eﬃciency of DNA enzymatic digestion. It is recommended refer to Step 7: Post-ampliﬁcation Clean-up to purify the nucleic acid samples using 2X NadPrep SP Beads, replace the elution reagent with Nuclease Free Water, and then proceed with library preparation.
